Paracrine Support

Paracrine support is a form of cell-to-cell communication in which one cell promotes the survival, function, or behavior of nearby cells through locally acting signals. Supporting cells release secreted molecules, such as growth factors, cytokines, or extracellular vesicles, which diffuse through the surrounding tissue and bind receptors on neighboring target cells, triggering specific intracellular responses. In biology, paracrine support helps coordinate tissue maintenance, development, immune activity, and repair without requiring direct cell contact. Studying these local signaling networks clarifies how cells cooperate within tissues and informs research on stem-cell culture, regenerative medicine, disease mechanisms, and therapeutic design.

Paracrine Signaling

Paracrine signaling allows cells to communicate with their immediate neighbors via secretion of signaling molecules. Such a signal can only trigger a response in nearby target cells because the signal molecules degrade quickly or are inactivated if not taken up. Prominent examples of paracrine signaling include nitric oxide signaling in blood vessels, synaptic signaling of neurons, the blood clotting system, tissue repair/wound healing, and local allergic skin reactions.Nitric oxide as a...

Here, we present a method using permeable membrane supports to facilitate the study of non-contact paracrine signaling used by tumor cells to suppress the immune response. The system is amenable to studying the role of tumor-secreted factors in dampening macrophage activation.

Proximal Culture System: An In Vitro Culture Technique to Study Paracrine Signaling Between Cells

In this video, we develop a proximal co-culture model of ovarian cancer cells and primary human mesothelial cells. This physiologically relevant model allows the study of paracrine signaling, where the localized concentrations of the secreted factors are maintained while preventing direct cellular contact.
