Reconstructed Human Epidermis

Reconstructed human epidermis is a laboratory-grown, three-dimensional tissue model that reproduces key structural and functional features of the outermost layer of human skin. Researchers typically culture human keratinocytes on a permeable support and expose the developing tissue to an air-liquid interface, which promotes cell proliferation, differentiation, and formation of stratified layers with a surface barrier. In biology, this model supports studies of epidermal development, skin barrier function, wound responses, and disease-related changes. It also provides a controlled platform for evaluating topical formulations, chemical irritation, and cellular mechanisms while reducing reliance on native human tissue.

In this report, we describe the three-dimensional skin reconstruct model which mimics human skin in architecture and composition. Melanocyte physiology, melanoma progression and the fate of dermal stem cells have been investigated using the skin reconstruct model. The model is also useful as a preclinical tool for drug assessment.
