Cell Cultivation Protocol

A Cell Cultivation Protocol is a standardized procedure for growing, maintaining, and handling cells under controlled laboratory conditions, enabling reproducible biological research. It typically combines aseptic technique with defined culture media, appropriate temperature and gas conditions, regular monitoring, and passaging or harvesting at suitable growth stages. In cancer research, these protocols support the maintenance of tumor-derived cell lines and primary cancer cells for studying proliferation, migration, signaling, drug responses, and treatment resistance. Consistent cultivation practices improve experimental comparability and help researchers generate reliable data for investigating cancer biology, evaluating therapeutic strategies, and developing clinically relevant models.

We present a simple method to construct 3D nematode cultivation systems called NGT-3D and NGB-3D. These can be used to study nematode fitness and behaviors in habitats that are more similar to natural Caenorhabditis elegans habitats than the standard 2D laboratory C. elegans culture plates.

2015

A novel reactor design, coined a high density bioreactor (HDBR), is presented for the cultivation and study of high density microbial communities. Here, the HDBR is successfully applied in a photobioreactor (PBR) configuration for the study of nitrogen metabolism by a mixed high density algal community.

Cultivation of Human Neural Progenitor Cells in a 3-dimensional Self-assembling Peptide Hydrogel

Here we describe the use of a self-assembling 3-dimensional scaffold to culture human neural progenitor cells. We present a protocol to release the cells from the scaffolds to be analysed subsequently e.g. by flow cytometry. This protocol might be adapted to other cell types to perform detailed mechanistically studies.
