Drug Screening Assay

A drug screening assay is a laboratory test that evaluates how candidate compounds affect a biological target, cell, or tissue, helping researchers identify molecules with therapeutic potential. In cancer research, cells or purified targets are exposed to controlled drug concentrations, and measured readouts such as viability, proliferation, apoptosis, or signaling activity are compared with untreated and control samples to determine potency and selectivity. These assays support hit identification, dose-response analysis, and prioritization of compounds for further study, including investigations of treatment resistance and combinations of anticancer agents. Reliable screening results can accelerate the development of safer, more effective cancer therapies.

This protocol describes the fabrication of elastic 3D macroporous microcryogels by integrating microfabrication with cryogelation technology. Upon loading with cells, 3D microtissues are generated, which can be readily injected in vivo to facilitate regenerative therapy or assembled into arrays for in vitro high-throughput drug screening.
