Organotypic Skin Reconstructs

Organotypic skin reconstructs are three-dimensional laboratory models that reproduce key structural and functional features of human skin, providing a more physiologically relevant system for cancer research than simple cell cultures. They are typically assembled from skin cells within an extracellular matrix and maintained under conditions that allow keratinocytes to stratify into an epidermis over a fibroblast-containing dermal equivalent. By incorporating tumor cells or patient-derived cancer samples, these models can reveal how malignant cells invade surrounding tissue, interact with the microenvironment, and respond to treatments. Organotypic skin reconstructs therefore support studies of tumor progression, therapeutic efficacy, and mechanisms of skin cancer development.

In this report, we describe the three-dimensional skin reconstruct model which mimics human skin in architecture and composition. Melanocyte physiology, melanoma progression and the fate of dermal stem cells have been investigated using the skin reconstruct model. The model is also useful as a preclinical tool for drug assessment.
