Pbmc-derived Cik Cells

PBMC-derived CIK cells are immune effector cells generated from peripheral blood mononuclear cells and expanded ex vivo for cancer research and immunotherapy studies. Their production typically combines interferon-γ priming, anti-CD3 stimulation, and interleukin-2-driven expansion, producing a heterogeneous population that includes CD3+CD56+ cells with natural killer-like, major histocompatibility complex-unrestricted cytotoxicity. These cells can recognize and kill a broad range of tumor targets through cytotoxic granules and death-receptor pathways, without requiring tumor-specific antigen matching. Researchers use PBMC-derived CIK cells to investigate antitumor mechanisms, evaluate combination treatments, and assess adoptive cell therapy strategies.

A Technique to Generate a Human PBMC-Engrafted Humanized Xenograft Mouse Model

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

We present a protocol to investigate the crosstalk between X-ray-irradiated Caco-2 and peripheral blood mononuclear cells (PBMC). The protocol starts with Caco-2 irradiation and set-up of the co-culture with PBMC; subsequently, trans-epithelial electrical resistance is measured regularly over 48 h and western blot performed in both Caco-2 and PBMC.

The ability of human embryonic stem cells to self-renew and differentiate into all cell types of the body suggests that they hold great promise for both medical applications and as a research tool for addressing fundamental questions in development and disease. This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
