Peptide Resolving Gel

Peptide resolving gel is a polyacrylamide electrophoresis matrix formulated to separate low-molecular-weight peptides and proteins that may migrate together in conventional gels. During SDS-PAGE, sodium dodecyl sulfate denatures samples and gives them a similar charge-to-mass ratio, while the gel’s tighter pore structure slows smaller molecules sufficiently to improve size-based resolution. In cancer research, this approach supports analysis of peptide fragments, signaling proteins, proteolytic products, and other small biomarkers in complex samples. Improved separation can strengthen downstream staining, band quantification, and immunoblot interpretation, helping researchers characterize molecular changes associated with tumor development and treatment response.

Here we present protocols for affinity purification of protein complexes and their separation by blue native PAGE, followed by protein correlation profiling using label free quantitative mass spectrometry. This method is useful to resolve interactomes into distinct protein complexes.
