Pharmacological Rescue

Pharmacological rescue is an experimental strategy that uses a drug or other bioactive compound to restore a cellular function or phenotype altered by genetic, chemical, or environmental perturbation. The approach works by modulating the affected molecular pathway, compensating for pathway disruption, or reversing a drug-induced response, after which researchers compare treated and untreated cells or model systems. In cancer research, pharmacological rescue can help validate therapeutic targets, clarify mechanisms of tumor-cell survival and drug resistance, and determine whether a specific pathway contributes to disease-associated phenotypes. These findings support more precise interpretation of gene-function studies and the development of rational combination treatments.

Capsulorhexis is an important step in phacoemulsification surgery. A surgeon creates a continous curvilinear tear on the anterior lens capsule by controlling the tearing vector forces. A peripherally extended tear is a serious complication. This video demonstrates a novel technique of rescuing capsular radial tear out using a cystotome.

A parasite-rescue and transformation assay with THP1 cells infected in vitro with Leishmania donovani has been optimized for anti-leishmanial drug screening. The assay involves differentiation of THP1 cells, infection with promastigotes, treatment with test drugs, controlled lysis of the infected macrophages, rescue of amastigotes, transformation to promastigotes and monitoring promastigote growth and proliferation with a fluorometric assay.

We describe a method for depletion-rescue experiments that preserves cellular integrity and protein homeostasis. Adenofection enables functional analyses of proteins within biological processes that rely on finely tuned actin-based dynamics, such as mitotic cell division and myogenesis, at the single-cell level.
