Submucosal Cell Injection

Submucosal cell injection is a technique that delivers a suspension of cells into the submucosal layer beneath an epithelial surface, creating a localized cellular model within tissue. Using a fine needle, researchers carefully penetrate the mucosa and deposit cells between the epithelium and deeper tissue, where cell survival, engraftment, and interactions with the surrounding microenvironment can be assessed. In cancer research, this approach supports localized tumor development and investigation of tumor–stroma interactions, invasion, and therapeutic responses. It can complement conventional culture and animal models by providing a controlled system for studying cancer behavior in a tissue-relevant context.

Here we demonstrate our protocol for isolation of basal and submucosal gland duct cells from mouse tracheas. We also demonstrate the method of injecting stem cells into the dorsal mouse fat pad to create an in vivo model of submucosal gland regeneration.

A New Ex Vivo Model for the Evaluation of Endoscopic Submucosal Injection Material Performance

We developed a new ex vivo model that applies constant tension to the porcine gastric specimen. This development made it possible to evaluate the performance (the height and duration of the submucosal elevation) of various SIMs accurately. The detailed setup methodology of this new model is explained.

This study reports a successful method of endoscopical adipose tissue-derived stromal cell (ADSC)-sheet transplantation for esophageal stricture prevention after an extended endoscopic submucosal dissection (ESD) in a swine model.

Spontaneous Metastasis Mouse Models: A Platform to Study the Metastatic Potential of Orthotopically Injected Colorectal Cancer Cells

This video describes the protocol to establish a spontaneous metastasis mouse model of colorectal cancer patient-derived xenografts or PDXs. These xenograft models partially mimic the features of human carcinomas and understand the critical aspects of human malignancies, including invasion and metastasis.
