Tumor Volume Analysis

Tumor volume analysis is the quantitative measurement of tumor size over time, providing an objective way to evaluate cancer growth and treatment response. Researchers estimate volume from physical dimensions, such as length and width measured with calipers, or from imaging data that captures tumor boundaries and calculates three-dimensional size. Repeated measurements generate growth curves and reveal changes associated with disease progression, therapeutic effects, or tumor regression. In cancer research, this analysis supports preclinical studies, enables comparisons among experimental groups, and helps assess the efficacy of anticancer treatments while reducing reliance on qualitative observations.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.

Segmentation of three-dimensional data from many imaging techniques is a major bottleneck in analysis of complex biological systems. Here, we describe the use of SuRVoS Workbench to semi-automatically segment volumetric data at various length-scales using example datasets from cryo-electron tomography, cryo soft X-ray tomography, and phase contrast X-ray tomography techniques.

Human tumor xenografts are vascularized and infiltrated by cells of mouse origin during the growth phase in vivo. To circumvent the bias caused by these contaminating cells during downstream analysis, we developed a method allowing for comprehensive depletion of all mouse cells by magnetic cell sorting.
