Combinatorial Library

A combinatorial library is a collection of diverse chemical compounds generated systematically from a defined set of building blocks, enabling rapid evaluation of many structures in a single research program. In chemistry, researchers create these libraries through parallel synthesis or split-and-mix methods, combining reactants in different sequences or proportions to produce distinct products, often on solid supports. After synthesis, the compounds are screened for properties such as biological activity, catalytic performance, binding affinity, or material behavior, using assays that identify promising members for further characterization. Combinatorial libraries therefore accelerate hit identification, structure-activity studies, and the development of pharmaceuticals, catalysts, and functional materials.

Peptide tertiary amides (PTAs) are a superfamily of peptidomimetics that include but are not limited to peptides, peptoids and N-methylated peptides. Here we describe a synthetic method which combines both split-and-pool and sub-monomer strategies to synthesize a one-bead one-compound library of PTAs.
