Depot Preparations

Depot preparations are long-acting drug formulations designed to release an active medicine gradually over an extended period, helping maintain therapeutic exposure after administration. In clinical practice, they typically form a drug reservoir at the administration site, where formulation properties and drug diffusion or dissolution control the rate of release and subsequent absorption into the circulation. This approach can reduce dosing frequency, support adherence, and provide more consistent drug levels than repeatedly administered immediate-release formulations. Depot preparations are used in areas such as hormone therapy, psychiatric care, contraception, and treatment of chronic conditions, where sustained drug delivery can improve continuity of care and therapeutic outcomes.

The method presented here uses 18F-Fluorodeoxyglucose (18F-FDG) positron emission tomography/computed tomography (PET-CT) and fat-water separated magnetic resonance imaging (MRI), each scanned following 2 hr exposure to thermoneutral (24 °C) and cold conditions (17 °C) in order to map brown adipose tissue (BAT) in adult human subjects.

Harvesting Peritoneal Fat Depots: A Technique to Study Ovarian Cancer Colonization of Peritoneal Adipose Tissues in Mouse Models

In this video, we use a euthanized female mouse model bearing ovarian cancer. We then describe a protocol to identify and extract fat depots or adipose tissues associated with the five primary peritoneal organs to study metastatic cancer cell colonization.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.

Due to the drastic and negative connection between obesity and other comorbidities, research on the role adipose plays in disease and overall health is warranted. We present a protocol for the isolation and excision of adipose depots allowing for the study of adipose using in situ and in vitro methods.
