5-mc Antibody Detection

5-mC antibody detection is an immunological method for identifying 5-methylcytosine, an epigenetic DNA modification that helps regulate gene activity without changing the DNA sequence. In this approach, antibodies selectively bind 5-mC residues in DNA, and the resulting antigen–antibody interaction is measured through a labeled detection signal in assays such as immunostaining, dot blotting, or ELISA. In developmental biology, the method helps track global or locus-associated changes in DNA methylation during embryogenesis, cell differentiation, and tissue formation. These measurements can reveal how epigenetic reprogramming contributes to developmental timing, lineage commitment, and abnormal developmental states.

The EpiMark 5-hmC and 5-mC Analysis Kit can be used to analyze and quantitate 5-methylcytosine and 5-hydroxymethylcytosine within a spe cific locus. The kit distinguishes 5-mC from 5-hmC by the addition of glucose to the hydroxyl group of 5-hmC via an enzymatic reaction utilizing β-glucosyltransferase (T4-BGT). When 5-hmC occurs In the context of CCGG, this modification converts a cleavable MspI site to a non-cleavable site.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.
