Fetal Vessel On-chip

Fetal vessel on-chip is a microfluidic model that recreates key features of developing blood vessels in a controlled laboratory environment, enabling researchers to study vascular development outside the embryo. The system typically combines endothelial cells within small perfused channels, where fluid flow generates shear stress and supports vessel formation, remodeling, and barrier function. In developmental biology, these platforms help examine how mechanical forces, cellular interactions, and molecular signals shape fetal vasculature. They can also support studies of placental and embryonic vascular disorders, drug responses, and developmental mechanisms that are difficult to investigate directly in living tissues.

Described here is a simple workflow to differentiate endothelial cells from human pluripotent stem cells followed by a detailed protocol for their mechanical stimulation. This allows for the study of the developmental mechanobiology of endothelial cells. This approach is compatible with downstream assays of live cells collected from the culture chip after mechanical stimulation.

Introduction to Organ-on-Chip Models Organ-on-chip models are advanced microphysiological systems that recreate key structural and functional features of human organs in a controlled laboratory setting. These devices integrate living human cells within microfluidic channels, allowing researchers to simulate tissue–tissue interfaces, fluid flow, and mechanical forces that are essential for normal organ function. Unlike traditional cell cultures, which often lack these dynamic physiological cues,...

Fetal and perinatal death is a common feature when studying genetic alterations affecting cardiac development. High-frequency ultrasound imaging has improved 2-D resolution and can provide excellent information on early cardiac development and is an ideal method to detect the impact on cardiac structure and function prior to death.
