Multiplex Short Tandem Repeat

Multiplex short tandem repeat (STR) analysis is a genetic method that examines several highly variable DNA regions simultaneously, producing informative profiles for distinguishing individuals and studying inheritance. It uses sets of locus-specific primer pairs to amplify multiple STR markers in a single polymerase chain reaction, after which fluorescently labeled products are separated by size and detected through capillary electrophoresis. Allele patterns can support human identification, paternity and kinship testing, forensic investigations, and population genetics. By combining information from many loci, multiplex STR analysis increases discriminatory power while reducing sample use, processing time, and laboratory effort.

Leprosy, caused by Mycobacterium leprae, is still endemic in many places. In order to learn about the spread and mode of transmission of leprosy, it is important to determine which strain of M. leprae has infected a patient. Variable numbers of tandem repeats (VNTR) typing is one such method.

The Multi-Locus Variable-number tandem-repeat Analysis (MLVA) assay presented here enables inexpensive, robust and portable high-resolution genotyping of the fish-pathogenic bacterium Yersinia ruckeri. Starting from pure cultures, the assay employs multiplex PCR and capillary electrophoresis to produce ten-loci MLVA profiles for downstream applications.
