Rifampicin Run-out

Rifampicin run-out is a bacterial genetics assay used to measure chromosome replication initiation and the number of active replication forks. Rifampicin blocks new transcription initiation, while replication forks that have already begun can continue until they complete the chromosome; cell division is often inhibited so completed chromosomes accumulate for measurement. Researchers commonly assess DNA content by flow cytometry to estimate chromosome equivalents, oriC-to-terminus relationships, replication timing, and fork number. The approach helps determine how genetic mutations, growth conditions, or replication-control factors affect chromosome duplication and genome stability.

The Xpert MTB/RIF test integrates sample decontamination, hands-free operation, on-board sample processing, and ultra-sensitive hemi-nested PCR for the simultaneous detection of Mycobacterium tuberculosis and rifampicin resistance, either in expectorated sputum or concentrated sputum sediments, in approximately two hours. Testing is standardized and requires only moderate laboratory infrastructure and training.
