Antibiotic Susceptibility Testing

Antibiotic susceptibility testing is a laboratory process that determines whether a bacterial pathogen is inhibited by specific antibiotics, helping guide effective treatment of infection. In methods such as disk diffusion and broth dilution, a standardized bacterial inoculum is exposed to selected drugs, and growth inhibition is measured to estimate the minimum inhibitory concentration or classify the isolate as susceptible, intermediate, or resistant using interpretive breakpoints. These results support antimicrobial selection, reveal resistance patterns, and help clinicians manage infections in patients with diverse immune responses. At the population level, susceptibility data also strengthen infection control, antimicrobial stewardship, and surveillance of emerging resistance.

We have developed a microfluidic platform for rapid antibiotic susceptibility testing. Fluid is passed at high speeds over bacteria immobilized on the bottom of a microfluidic channel. In the presence of stress and antibiotic, susceptible strains of bacteria die rapidly. However, resistant bacteria can survive these stressful conditions.

(2022)This video demonstrates the application of stimulated Raman scattering microscopy to assess susceptibility to antibiotics by tracking deuterium-labeled metabolic activity. By monitoring carbon–deuterium signals, the method enables rapid detection of resistant and susceptible bacterial populations.

A rapid bacterial pellet preparation from a positive blood culture can be used as a sample for applications such as identification by MALDI-TOF, Gram staining, antibiotic susceptibility testing and PCR-based test. The results can be rapidly communicated to clinicians to improve the outcome of patients suffering from bloodstream infections.
