Antibody Isotypes

Antibody isotypes are classes of immunoglobulins that share antigen-binding principles but differ in structure, distribution, and immune function. The major human isotypes, IgM, IgD, IgG, IgA, and IgE, arise when B cells undergo class-switch recombination, replacing the antibody heavy-chain constant region while preserving antigen specificity; cytokines and signals from helper T cells help direct this process. These differences determine whether antibodies activate complement, cross the placenta, protect mucosal surfaces, or recruit cells involved in allergic and antiparasitic responses. Understanding antibody isotypes helps explain infection control, vaccine-induced immunity, autoimmune disease, allergy, and the interpretation of serological tests.

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

Antibody Binding Specificity for Kappa (Vκ) Light Chain-containing Human (IgM) Antibodies: Polysialic Acid (PSA) Attached to NCAM as a Case Study

We present a protocol to identify protein moieties containing antigens for human and mouse IgM antibodies with specific kappa (Vκ) light chains. This protocol is not limited to IgM class antibodies but applies to all immunoglobulin isotypes that target their antigen with sufficiently high affinity during immunoprecipitations.

C57BL/6 mice have been used to study Hc pathogenesis and provide the best model. We are exploring the potential benefits of humoral immunity against this fungus and generated several mAbs [to histone H2B and a heat shock protein 60kDa] that we tested for their protective efficacy after intraperitoneal administration.

A Technique to Assess the In Vivo Localization of Tumor-Specific Antibodies

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.
