Asymmetric Primer Concentration

Asymmetric primer concentration is a PCR design in which one primer is supplied at a higher concentration than its counterpart to preferentially produce single-stranded DNA. During amplification, the limiting primer becomes depleted after early cycles, while the excess primer continues copying the template strand, shifting the reaction from balanced double-stranded products toward one-strand products. In immunology and infection research, this approach can generate single-stranded DNA templates for hybridization probes, sequencing, or binding assays, supporting the detection and characterization of pathogen-derived or immune-related nucleic acids.

Here, we present a protocol to quantify precise stepping in rodents. Cortical and the spinal central pattern generator signals are required for precise foot-placement during obstructed locomotion. We report here the novel constrained walking task that directly examines precise stepping behavior.

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.
