Bacterial Load Analysis

Bacterial load analysis is the measurement of the number of bacteria present in a biological or environmental sample, providing a quantitative view of infection severity and microbial growth. It commonly relies on serial dilution and culture to count colony-forming units, or on quantitative PCR to estimate bacterial DNA through amplification of target sequences. In immunology and infection research, these measurements help assess pathogen replication, host immune control, and responses to antimicrobial treatment. Comparing bacterial loads across time points or experimental groups supports evaluation of disease progression, treatment efficacy, and mechanisms of host-pathogen interaction.

Bacterial Infection Quantitation Assay: A Method to Quantify Intraocular Bacterial Load in Mouse Model of Bacterial Endophthalmitis

2025

This video describes the technique of quantifying bacterial infection from eye homogenates obtained from a murine model of bacterial endophthalmitis caused by Bacillus cereus. This technique helps study the host immune response and its dependency on the host and ocular bacterial gene expression in murine models.

Source: Ahn, J. J., et al. Experimental Infection with Listeria monocytogenes as a Model for Studying Host Interferon-γ Responses. J. Vis. Exp. (2016)The video demonstrates bacterial load measurement in spleen and liver tissues isolated from mice infected with an intracellular bacterial pathogen. Bacteria, accumulated within tissue-resident phagocytes during systemic infection, are released using detergent-based lysis and mechanical disruption. Serial dilutions of the homogenates are plated on...

2011

Listeria monocytogenes is a model organism for studying immune responses and genetic susceptibility to intracellular bacteria in mice. This method enables one to measure bacterial load and generate single-cell suspensions of the liver and spleen from mice for FACS analysis to determine changes in immune cells due to Listeria infection.

Quantification of Tissue Bacterial Load in a Mouse Model of Chronic Intestinal Infection

Source: Lo, B. C., et al. Chronic Salmonella Infection Induced Intestinal Fibrosis. J. Vis. Exp. (2019)The video demonstrates the quantification of bacterial load in a mouse model of chronic infection with streptomycin-resistant Salmonella typhimurium. Following euthanasia and sterile dissection, cecal and splenic tissues are harvested, homogenized with steel beads to release bacteria, and serially diluted. Dilutions are plated on selective agar, and visible colonies are counted to estimate...
