Anaerobic Incubation

Anaerobic incubation is the cultivation of microorganisms under conditions that exclude oxygen, an essential approach for studying bacteria unable to grow in air. It works by placing inoculated cultures in sealed anaerobic chambers, jars, or bags containing oxygen-removing systems, often with a reducing atmosphere that limits oxygen exposure during growth. In immunology and infection research, anaerobic incubation supports the isolation and identification of obligate anaerobes from clinical specimens, analysis of polymicrobial communities, and investigation of host-microbe interactions in oxygen-limited tissues. Reliable oxygen control improves culture recovery and helps clarify how anaerobic pathogens contribute to disease, antimicrobial responses, and microbiome-associated immune regulation.

Clostridium difficile is a pathogenic bacterium that is a strict anaerobe and causes antibiotic associated diarrhea (AAD). Here, methods for isolating, culturing and maintaining C. difficile vegetative cells and spores are described. These techniques necessitate an anaerobic chamber, which requires regular maintenance to ensure proper conditions for optimal C. difficile cultivation.

Visualization of Internalized Anaerobic Bacteria within Host Endothelial Cells

In this video, we demonstrate a procedure to visualize internalized Porphyromonas gingivalis, an anaerobic pathogenic bacterium, within human endothelial cells using confocal microscopy. The bacteria are stained with a fluorescent dye while the host endothelial cell actin cytoskeleton is labeled with a different fluorescent dye. The internalized Porphyromonas gingivalis inside endothelial cells are visualized using confocal microscopy.
