Hepatic Stellate Cells

Hepatic stellate cells are specialized perisinusoidal cells in the liver that store vitamin A and help maintain tissue structure, making them central to liver repair and disease. In a healthy liver, they remain quiescent with lipid droplets, but injury, inflammatory cytokines, or infection can activate them into myofibroblast-like cells that produce extracellular matrix and reshape the hepatic microenvironment. Their interactions with hepatocytes, Kupffer cells, endothelial cells, and lymphocytes influence inflammation, fibrogenesis, and pathogen responses. Studying hepatic stellate cells therefore supports research on viral and parasitic infections, chronic liver disease, immune regulation, antifibrotic therapies, and host-directed treatment strategies.

Here we describe a method for the isolation of hepatic stellate cells from mouse liver. For stellate cell purification, mouse livers are digested in situ and in vitro by pronase-collagenase treatment prior to density gradient centrifugation. This technique yields highly pure hepatic stellate cells.

Primary hepatocytes provide a valuable tool to evaluate biochemical, molecular, and metabolic functions in a physiologically relevant experimental system. We describe a reliable protocol for rat in situ liver perfusion, which consistently generates viable hepatocytes up to 1.0 × 108 cells per preparation with cell viability between 88 ~ 96%.

Source: Meister, T. L., et al. A Cell Culture Model for Producing High Titer Hepatitis E Virus Stocks. J. Vis. Exp. (2020).This video demonstrates the process of isolating hepatitis E virus (HEV) particles from transfected human hepatocellular carcinoma cells. Extracellular viral particles are collected from filtered supernatants, while intracellular particles are released by subjecting the cells to repeated freeze-thaw cycles. The lysate is centrifuged, and the supernatant containing HEV...
