Islet Autoantibodies

Islet autoantibodies are immune proteins that recognize molecules in the pancreatic islets, especially antigens associated with insulin-producing beta cells, and they are important markers of autoimmune diabetes. They arise when immune tolerance to islet components breaks down, generating antibodies against targets such as insulin, glutamic acid decarboxylase, IA-2, or ZnT8; although these antibodies may not directly destroy beta cells, their presence reflects an ongoing autoimmune response. Testing for islet autoantibodies supports the diagnosis and classification of type 1 diabetes, helps identify individuals at elevated risk before symptoms appear, and provides insight into the development and progression of beta-cell autoimmunity.

Here, we presented the methods to detect human islet autoantibodies using electrochemiluminescence (ECL) assays. The protocol, used to predict type 1 diabetes, can be expanded upon to detect autoantibodies for other autoimmune diseases.

An Assay to Detect Autoantibodies in Human Serum using a Hippocampal Neuronal Culture

This video demonstrates a technique for identifying antibodies against specific antigens in a sample using a rat hippocampal neuronal culture. By exposing cultured neurons to patient samples containing autoantibodies targeting N-Methyl-D-aspartate receptors (NMDARs), the presence of autoantibodies bound to the neuronal surface is revealed through immunostaining.

In the animal model described in our present work, purified IgG antibodies against a stretch of 200 amino acids (aa 757-967) of collagen VII are injected repeatedly into mice reproducing the blistering phenotype as well as the histo- and immunopathological features characteristic to human epidermolysis bullosa acquisita (EBA)1.

Pancreatic Islets Isolation through Collagenase Perfusion: An Enzymatic Method to Isolate Pancreatic Islet Cells

2023

Pancreatic islets, also called the Islets of Langerhans, are a cluster of endocrine cells which produces hormones for glucose regulation and other important biological functions. This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.
