Liver Capsular Macrophages

Liver capsular macrophages are innate immune cells positioned within the connective-tissue capsule covering the liver, where they provide an early defense against invading microbes and help regulate local inflammation. They detect pathogen-associated molecular patterns through pattern-recognition receptors, then engulf particles and release cytokines and chemokines that coordinate recruitment of other immune cells. Unlike resident macrophages deeper in the liver, their surface location enables rapid surveillance of the organ boundary and nearby tissues. Studying liver capsular macrophages in immunology and infection clarifies how the liver responds to pathogens, contributes to inflammatory disease, and may guide macrophage-targeted strategies for controlling infection or limiting tissue damage.

A novel method to obtain macrophages from primary culture of rat liver cells is described. This method utilizes the proliferation of macrophages in the culture, followed by shaking of culture flasks and purification by selective attachment to plastic dishes. This technique efficiently provides liver macrophages without complex equipment and skills.

This manuscript describes a modeling technique of capsular infarct. Here we utilized a modified photothrombotic technique with low intensity of light after pre-surgery target mapping. Using this technique, we created a circumscribed capsular infarct model with persistent motor impairment.

Drug-Induced Senescence in Liver Cells Promotes M2 Macrophage Polarization: Implications for Tyrosine Kinase Inhibitor-Associated Hepatotoxicity

Here, we study hepatotoxicity caused by lapatinib and neratinib through SASP-driven macrophage polarization, which offers a deeper mechanistic understanding beyond traditional toxicity tests. This approach demonstrates how senescent hepatocytes influence immune responses. This is particularly valuable for assessing long-term drug effects and discovering new therapeutic targets for managing drug-induced hepatotoxicity.
