Parafilm Gasket

A Parafilm gasket is a flexible film seal used to close the junction between a container, lid, plate, or other laboratory components, helping maintain controlled experimental conditions. When stretched over an opening or compressed between contacting surfaces, Parafilm conforms to small irregularities and forms a temporary barrier that limits evaporation, spills, and entry of airborne contaminants. In immunology and infection research, these gaskets can help stabilize samples and reagents during incubation, transport, or storage, including workflows involving microplates, cultures, and diagnostic assays. Their low cost, easy removal, and adaptability make them useful for routine containment, although they do not replace validated sterile closures.

An In Vitro Flow Investigation of the Effect of Stromal Cells on Leukocyte Recruitment

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.

We illustrate here an in vitro membrane binding assay in which interactions between HIV-1 Gag and lipid membranes are visually analyzed using YFP-tagged Gag synthesized in a wheat germ-based in vitro translation system and GUVs prepared by an electroformation technique.

Recording Multicellular Behavior in Myxococcus xanthus Biofilms using Time-lapse Microcinematography

To study Myxococcus xanthus swarm behavior, we have designed a time-lapse microcinematography protocol that can be modified for different assays. It employs standard growth conditions adapted for microscopy, and yields reproducible results by the use of inexpensive, reusable silicone gaskets. We have used this method to quantify multicellular chemotaxis.

Reconstituting functional membrane proteins into giant liposomes of defined composition is a powerful approach when combined with patch-clamp electrophysiology. However, conventional giant liposome production may be incompatible with protein stability. We describe protocols for producing giant liposomes from pure lipids or small liposomes containing ion channels.
