Viral Gene Activation

Viral gene activation is the process by which a virus initiates transcription of its genetic information in an infected cell, enabling replication and manipulation of host responses. It typically begins when viral genomes reach the nucleus or cytoplasm and viral or host transcription factors bind regulatory sequences, while signaling pathways and chromatin state determine which genes turn on first. Early gene expression can produce proteins that replicate the genome, evade innate immunity, or regulate later viral programs. In immunology and infection research, understanding these controls helps explain viral persistence, pathogenesis, latency, and opportunities for antiviral or vaccine development.

We describe the usage of a fluorescent reporter vaccinia virus that enables real-time measurement of viral infectivity and gene expression through the stage-specific expression of spectrally distinct reporter fluorophores. We detail a plate-based method for accurately identifying the stage at which virus replication is affected in response to small molecule inhibition.
