Viral Infectivity Assay

A viral infectivity assay is a laboratory method that measures the ability of virus particles to infect susceptible cells and produce new infectious progeny, rather than simply detecting viral genetic material. In a typical assay, researchers expose cultured cells to serial virus dilutions, allow infection to proceed, and quantify plaques, infected cell foci, or cytopathic effects to calculate infectious titer. These measurements support the characterization of viral replication, neutralizing antibody activity, vaccine-induced immunity, and antiviral efficacy. In immunology and infection research, infectivity assays help distinguish functional changes in virus-host interactions from changes in particle number alone.

An In Vitro Assay to Examine the Antibody-Dependent Enhancement of Viral Infection

This video demonstrates an assay to measure antibody-dependent enhancement (ADE) of Dengue virus infection using Zika Virus Convalescent Serum. The assay uses Dengue reporter viral particles (RVPs) encoding a green fluorescent protein. Serum antibodies against the Zika Virus cross-react with RVPs to form a complex. The bound antibodies enhance RVP uptake by cells expressing the Fc receptor, which is detected using flow cytometry.
