Antibody Expression

Antibody expression is the production of immunoglobulin proteins by B cells or engineered host cells, enabling researchers to obtain antibodies for studying and controlling immune responses. In vivo, activated B cells differentiate into plasma cells that transcribe immunoglobulin genes and secrete assembled heavy- and light-chain proteins; in biotechnology, cloned antibody genes are introduced into systems such as mammalian cells, which fold, assemble, and modify the resulting molecules. Expressed antibodies support pathogen detection, antigen characterization, neutralization studies, diagnostics, and therapeutic development. In immunology and infection research, expression analysis also helps connect antibody structure and abundance with protective or dysregulated immune responses.

Controlling protein expression is not only essential to every organism alive, but also an important strategy to investigate protein functions in cellular models. The protocol presented shows the application of antibody interference in mammalian cells including primary hippocampal neurons and demonstrates the use of three-dimensional reconstructions in studying protein function.

Antibody Microarray: A Technique to Study the Protein Expression of miRNA Treated Lung Cancer Cells

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
