Ex Vivo Drug Testing

Ex vivo drug testing is the evaluation of therapeutic compounds in living cells, tissues, or organs removed from an organism, providing a controlled model that retains aspects of native biology. In this approach, freshly isolated or preserved samples are maintained under laboratory conditions, exposed to defined drug concentrations, and assessed for responses such as molecular changes, tissue function, efficacy, or toxicity. In medicine, ex vivo testing supports drug screening, disease modeling, and personalized treatment decisions by revealing how patient-derived material may respond before clinical administration. It also complements in vitro systems and animal studies, helping refine experimental design and guide translational research.

Ex vivo ERG can be used to record electrical activity of retinal cells directly from isolated intact retinas of animals or humans. We demonstrate here how common in vivo ERG systems can be adapted for ex vivo ERG recordings in order to dissect the electrical activity of retinal cells.

Patient-derived xenografts of glioblastoma multiforme can be miniaturized into living microtumors using 3D human biogel culture system. This in vivo-like 3D tumor assay is suitable for drug response testing and molecular profiling, including kinomic analysis.

This manuscript describes the development of an animal model that allows for the direct testing of the effects of tumor hypoxia on metastasis and the deciphering the mechanisms of its action. Although the experiments described here focus on Ewing sarcoma, a similar approach can be applied to other tumor types.
