Human Cardiomyocytes

Human cardiomyocytes are specialized heart muscle cells that generate the electrical signals and coordinated contractions required to pump blood throughout the body. Their function depends on action potentials that regulate calcium entry, triggering actin–myosin interactions and sarcomere shortening, while cell-to-cell connections synchronize contraction across cardiac tissue. In medicine, human cardiomyocytes support research into cardiac development, arrhythmias, myocardial injury, and inherited heart disease. Cardiomyocytes derived from human pluripotent stem cells also provide models for drug screening, cardiotoxicity testing, and regenerative strategies, helping researchers evaluate treatments under more physiologically relevant conditions.

Pluripotent stem cells, either embryonic or induced pluripotent stem (iPS) cells, constitute a valuable source of human differentiated cells, including cardiomyocytes. Here, we will focus on cardiac induction of iPS cells, showing how to use them to obtain functional human cardiomyocytes through an embryoid bodies-based protocol.

The cardiac biowire platform is an in vitro method used to mature human embryonic and induced pluripotent stem cell-derived cardiomyocytes (hPSC-CM) by combining three-dimensional cell cultivation with electrical stimulation. Current knowledge on the cellular basis of cardiac diseases mostly relies on studies on animal models. Here we describe and validate a novel method to obtain single viable cardiomyocytes from small surgical samples of human ventricular myocardium. Human ventricular myocytes can be used for electrophysiological studies and drug testing.

Efficient Derivation of Human Cardiac Precursors and Cardiomyocytes from Pluripotent Human Embryonic Stem Cells with Small Molecule Induction

We have established a protocol for induction of cardioblasts direct from pluripotent human embryonic stem cells maintained under defined conditions with small molecules, which enables derivation of a large supply of human cardiac progenitors and functional cardiomyocytes for cardiovascular repair.
