Longitudinal Imaging

Longitudinal imaging is the repeated acquisition and comparison of medical images from the same patient over time, enabling clinicians and researchers to monitor biological change rather than evaluate a single time point. By using consistent imaging methods, anatomical landmarks, quantitative measurements, or imaging biomarkers can be aligned across examinations to identify disease progression, treatment response, or recovery. In medicine, longitudinal imaging supports diagnosis, prognosis, therapy planning, and follow-up for conditions that evolve over days, months, or years. It also strengthens clinical research by revealing temporal patterns and measuring outcomes in individual patients and study populations.

Sometimes we want to see how people change over time, as in studies of human development and lifespan. When we test the same group of individuals repeatedly over an extended period of time, we are conducting longitudinal research. Longitudinal research is a research design in which data-gathering is administered repeatedly over an extended period of time. For example, we may survey a group of individuals about their dietary habits at age 20, retest them a decade later at age 30, and then again...

An approach is here presented for long-term intravital imaging using optically clear, silicone windows that can be glued directly to the tissue/organ of interest and the skin. These windows are cheaper and more versatile than others currently used in the field, and the surgical insertion causes limited inflammation and distress to the animals.

This manuscript describes a procedure to track the remodeling of the cerebrovasculature during amyloid plaque accumulation in vivo using longitudinal two-photon microscopy. A thinned-skull preparation enables the visualization of fluorescent dyes to assess the progression of cerebrovascular damage in a mouse model of Alzheimer's disease.
