Lymphocyte Function

Lymphocyte function describes how specialized white blood cells recognize threats, coordinate immune responses, and establish protection against disease. B lymphocytes produce antibodies after activation, while T lymphocytes use antigen receptors to identify infected or abnormal cells, regulate other immune cells, or destroy targets; memory lymphocytes support faster responses during subsequent exposure. Understanding these coordinated processes is central to medicine because altered lymphocyte activity contributes to immunodeficiency, autoimmune disease, infection, cancer, and transplant rejection. Laboratory studies of lymphocyte activation, signaling, proliferation, and cytotoxicity help evaluate immune status, guide diagnosis, and inform vaccines, immunotherapies, and other treatments.

Radial Mobility and Cytotoxic Function of Retroviral Replicating Vector Transduced, Non-adherent Alloresponsive T Lymphocytes

We describe a protocol to monitor radial mobility of non-adherent immune cells in vitro using a cell sedimentation manifold/slide apparatus. Cell migration is tracked on monolayers of tumor cells or on extracellular matrix proteins. Examination by light and fluorescence microscopy allows for observation of cell mobility and cytotoxic functionality.

Lymphocytes are the major players in adaptive immune responses. Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.
