Macropinocytic Cups

Macropinocytic cups are transient, actin-rich membrane ruffles that extend around and engulf large volumes of extracellular fluid, forming macropinosomes through macropinocytosis. Their formation begins when signaling pathways activate actin polymerization at the plasma membrane, driving outward protrusion; cup closure then traps fluid, soluble molecules, and sometimes particulate material for intracellular processing. In medicine, macropinocytic cups are relevant to nutrient acquisition by cancer cells, antigen uptake by immune cells, and pathogen entry, making them important for understanding tumor metabolism, infection, and immune regulation. Imaging and molecular analysis of cup formation can help identify therapeutic targets and clarify disease mechanisms.

This protocol describes an approach for in toto labeling and multidimensional imaging of zebrafish early eye development. We describe labeling, embedding, and four dimensional (4D) imaging using laser scanning confocal microscopy, and considerations for optimizing acquisition of datasets for dissecting mechanisms of optic cup morphogenesis.

Synthesis and Functionalization of Nitrogen-doped Carbon Nanotube Cups with Gold Nanoparticles as Cork Stoppers

We discussed the synthesis of individual graphitic nanocups using a series of techniques including chemical vapor deposition, acid oxidation and probe-tip sonication. By citrate reduction of HAuCl4, the graphitic nanocups were effectively corked with gold nanoparticles due to the chemically reactive edges of the cups.
