Matrigel Embedding

Matrigel embedding is a three-dimensional cell-culture technique in which cells, tissues, or organoids are suspended in Matrigel, a basement-membrane extract that supports tissue-like growth and organization. Kept liquid at low temperature and warmed to physiological conditions, Matrigel polymerizes into a protein-rich gel containing components such as laminin and collagen IV, creating a scaffold for cell adhesion, polarity, and signaling. In medicine and biomedical research, this approach enables organoid formation, tumor invasion studies, disease modeling, and drug-response testing, while helping investigators examine how cells interact with an extracellular matrix in controlled experimental settings.

This video demonstrates how to maintain the growth of human embryonic stem cells (hESCs) in feeder cell-free conditions and how to continuously passage hESCs in feeder cell-free conditions. Confirmation of hESC pluripotency grown in feeder cell-free conditions by immunofluorescence microscopy is also demonstrated. Part 3 of 3.
