Muscle Volume Measurement

Muscle volume measurement is the quantitative assessment of skeletal muscle size, an important indicator of body composition, physical function, and changes associated with disease or treatment. In medicine, clinicians and researchers estimate volume by acquiring anatomical images, such as magnetic resonance imaging or computed tomography scans, separating muscle tissue from surrounding structures through image segmentation, and calculating the enclosed volume across sequential slices. These measurements support the evaluation of muscle wasting, rehabilitation, neuromuscular disorders, aging, and responses to exercise or therapy. Standardized imaging-based assessment can also improve clinical monitoring and provide objective outcomes for research studies.

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.
