Nose-only Exposure

Nose-only exposure is a controlled inhalation method in which an individual breathes an aerosol, vapor, gas, or particulate substance through the nose while other exposure routes are minimized. The subject is positioned so the nose receives a measured atmosphere, allowing researchers to regulate concentration, exposure duration, airflow, and inhaled dose while limiting contamination of the skin or fur. In medicine and biomedical research, this approach supports studies of respiratory toxicology, drug delivery, airway inflammation, and dose-response relationships. It can help clarify how airborne substances enter and affect the respiratory tract, although careful monitoring is needed to account for stress, breathing patterns, and exposure uniformity.

The inhalation unit described herein can generate, sample for characterization, and uniformly deposit a drug aerosol in the lungs of rodents. This enables the pre-clinical determination of the efficacy and safety of drug doses deposited in the lungs; key data enabling clinical inhaled drug development.

A nose-only inhalation toxicity chamber capable of testing inhalation toxicity at four different exposure concentrations was designed and validated for flow field uniformity and cross-contamination between the exposure ports for each concentration. Here, we present a protocol to confirm that the designed chamber is effective for inhalation toxicity testing.

Fruit Volatile Analysis Using an Electronic Nose

A rapid method for volatile compound analysis in fruit is described. The volatile compounds present in the headspace of a homogenate of the sample are rapidly separated and detected with ultra-fast gas chromatography (GC) coupled with a surface acoustic wave (SAW) sensor. A procedure for data handling and analysis is also discussed.
