Organ Ex Vivo Imaging

Organ ex vivo imaging is the visualization and analysis of an organ after surgical removal, enabling researchers to examine anatomy, cellular activity, and disease-related changes outside the body. During imaging, the organ is placed in a controlled environment and may be maintained with oxygenated media or perfusion to preserve tissue structure and function while optical, fluorescent, or other imaging methods capture biological features. In medicine, this approach supports studies of pathology, treatment response, organ preservation, and transplantation, while linking macroscopic tissue changes with underlying cellular processes. It also provides a valuable bridge between laboratory models and clinical imaging.

Mesoscopic fluorescence tomography operates beyond the penetration limits of tissue-sectioning fluorescence microscopy. The technique is based on multi-projection illumination and a photon transport description. We demonstrate in-vivo whole-body 3D visualization of the morphogenesis of GFP-expressing wing imaginal discs in Drosophila melanogaster.

Optical clearing techniques, such as absorbing molecules solutions, reduce light scattering temporarily to enhance biological imaging and offer biocompatible, reversible in vivo imaging strategies for deeply embedded organs. This protocol presents detailed experimental procedures as well as advanced image processing to achieve high-quality, dynamic imaging for real-time dynamics in living animals.

Plant viral nanoparticles (VNPs) are promising platforms for applications in biomedicine. Here, we describe the procedures for plant VNP propagation, purification, characterization, and bioconjugation. Finally, we show the application of VNPs for tumor homing and imaging using a mouse xenograft model and fluorescence imaging.

An Ex Vivo Gut Organ Culture System to Study Host-Microbiota Interactions

In this video, we describe the methodology for setting up an ex vivo gut organ culture system to study the interactions between mouse intestinal tissue fragments and specific gut bacteria. The device consists of two pairs of input-output ports, the first connecting the intestinal tissue lumen and supplying the specific bacterial suspension into it, and the second continuously supplying media to the device's wells to maintain ex vivo tissue viability.
