Integrin Blocking Antibodies

Integrin blocking antibodies are therapeutic or experimental antibodies that bind cell-surface integrins and interfere with these adhesion receptors’ interactions with extracellular-matrix proteins or other cell-surface ligands. By occupying integrin-binding sites or stabilizing an inactive receptor conformation, they can reduce outside-in signaling, cell adhesion, migration, and activation; effects depend on the targeted integrin and tissue context. In medicine, these antibodies help investigate leukocyte trafficking, angiogenesis, platelet aggregation, and tumor-cell behavior, while clinically developed agents can modulate immune-cell movement or platelet function in conditions including inflammatory disease and thrombosis.

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling." In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

In this study, we describe an improved protocol for a multiplexed high-throughput antibody microarray with lectin detection method that can be used in glycosylation profiling of specific proteins. This protocol features new reliable reagents and significantly reduces the time, cost, and lab equipment requirements as compared to the previous procedure.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
