Rpe Degeneration

RPE degeneration is the progressive loss or dysfunction of retinal pigment epithelial cells, which support photoreceptors and maintain vision. These cells normally phagocytose shed photoreceptor outer segments, regulate nutrient exchange across the blood-retina barrier, and protect the retina from oxidative stress; when these functions fail, photoreceptors become damaged and may die. RPE degeneration is a central feature of age-related macular degeneration and several inherited retinal diseases, making it an important focus of medical research. Studying its cellular mechanisms supports the development of biomarkers, protective therapies, gene-based treatments, and cell-replacement strategies aimed at preserving or restoring vision.

Dissection of Human Retina and RPE-Choroid for Proteomic Analysis

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
