Spontaneous Metastasis

Spontaneous metastasis is the natural spread of cancer cells from a primary tumor to distant organs without experimentally introducing tumor cells into the circulation. It develops through sequential steps that include local invasion, intravasation into blood or lymphatic vessels, survival during transit, extravasation, and colonization of a new tissue. In medicine and cancer research, spontaneous metastasis models allow investigators to study these events in the context of primary tumor growth and interactions with the surrounding microenvironment. They support evaluation of metastatic mechanisms, disease progression, therapeutic responses, and strategies designed to prevent or treat secondary tumors.

An orthotopic breast cancer primary tumor model and surgical removal of primary tumor to extend mouse life to generate spontaneous metastasis are described. The tumor growth and progression are monitored and quantified by luciferase fluorescence imaging.
