Tear Fluid Sampling

Tear fluid sampling is a minimally invasive technique for collecting tears to measure biological molecules and assess ocular and systemic health. Samples may be obtained from the tear meniscus with a capillary tube or absorbed onto a strip; the collected fluid is then eluted or analyzed directly using biochemical, immunological, or molecular assays. Because tears contain proteins, lipids, electrolytes, metabolites, and shed cellular material, their composition can reflect inflammation, infection, dry-eye dysfunction, and other physiological changes. Standardizing collection volume, sampling location, timing, and contamination control improves reproducibility, supporting noninvasive biomarker research, clinical assessment, and the development of point-of-care tests.

2025

This protocol describes a unique clinical protocol for collecting human tear fluid samples using phenol red threads and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based workflow to discover the tear lipidomic profile. The simple methyl tert-butyl ether (MTBE)/methanol biphasic separation method enables rapid tear lipid extraction with high recovery for tear biomarker discovery.

A method for mass spectrometric analysis of endogenous peptides in human cerebrospinal fluid (CSF) is presented. By employing molecular weight cut-off filtration, chromatographic pre-fractionation, mass spectrometric analysis and a subsequent combination of peptide identification strategies, it was possible to expand the known CSF peptidome nearly ten-fold compared to previous studies.
