Extracellular Vesicle Analysis

Extracellular vesicle analysis is the study of membrane-bound particles released by cells to determine their size, composition, abundance, and biological function. In neuroscience, researchers isolate vesicles from biofluids or cell cultures, then characterize their lipid membranes, proteins, and nucleic acid cargo using complementary separation and detection methods. Because extracellular vesicles can transport molecular signals between neurons, glia, and other cells, their contents may reveal mechanisms of neural communication, inflammation, and disease progression. Analysis supports biomarker discovery, evaluation of neuronal and glial responses, and development of minimally invasive approaches for studying neurological disorders.

Many different methods exist for the measurement of extracellular vesicles (EVs) using flow cytometry (FCM). Several aspects should be considered when determining the most appropriate method to use. Two protocols for measuring EVs are presented, using either individual detection or a bead-based approach.

This protocol details a method to isolate extracellular vesicles (EVs), small membranous particles released from cells, from as little as 10 μl serum samples. This approach circumvents the need for ultracentrifugation, requires only a few minutes of assay time, and enables the isolation of EVs from samples of limited volumes.
