Glutamatergic Neuron Differentiation

Glutamatergic neuron differentiation is the biological process by which neural progenitor cells acquire the identity and function of neurons that use glutamate as their primary excitatory neurotransmitter. It is guided by coordinated changes in gene expression and developmental signaling, which establish neuronal morphology, synaptic machinery, glutamate production, and selective neurotransmitter release. In neuroscience research, this process is modeled using stem cell cultures, organoids, and induced pluripotent stem cells to study cortical development and circuit formation. These models also support investigations of epilepsy, autism, neurodegenerative disease, and potential therapies by providing human glutamatergic neurons for disease modeling and drug evaluation.

This video showcases a technique for co-culturing glutamatergic neurons and pediatric high-grade glioma (pHGG) cells within a microfluidic device. Human induced pluripotent stem cells are introduced into the microfluidic system and incubated to promote their differentiation into mature cortical glutamatergic neurons. Following this, the pHGG cells are introduced onto the mature neurons, establishing a co-culture and enabling interactions between neurons and pHGG cells.
