Honeybee Learning

Honeybee learning is the process by which honeybees acquire and retain associations between sensory cues, actions, and meaningful outcomes such as food, helping researchers study the neural basis of behavior. In classical conditioning experiments, a bee can learn to associate an odor or color with a sucrose reward, producing a measurable proboscis extension response; changes in neural circuits, particularly within the mushroom bodies, support memory formation. Neuroscience studies of honeybee learning provide insight into sensory processing, associative memory, decision-making, and neural plasticity. Their tractable behavior and compact nervous system make them valuable models for comparing cognition across species.

Behavioural Pharmacology in Classical Conditioning of the Proboscis Extension Response in Honeybees (Apis mellifera)

We demonstrate how to implement a behavioral pharmacology method in an appetitive olfactory conditioning paradigm in honeybees (Apis mellifera) by systemic application of drugs. This method allows investigation of the mechanisms underlying learning and memory formation in a simple and reliable way.

Simultaneous Long-term Recordings at Two Neuronal Processing Stages in Behaving Honeybees

Simultaneous extracellular long term recordings from two different brain neuropiles or two different anatomical tracts were established in honeybees. These recordings allow the investigation of temporal aspects of neuronal processing across different brain areas at the single neuron as well as at the ensemble level in a behaving animal.

Here we describe our detailed protocol for the preparation of single-cohort honeybee colonies – a useful tool for analyzing the role-associated worker physiology. We also describe detailed protocols for treating workers with juvenile hormone and ecdysone to evaluate the involvement of these hormones in the regulation of worker behavior and/or physiology.

Bees can be conditioned in an appetitive olfactory learning paradigm (PER-conditioning). Using odors as stimuli, we established a method in which behavior is recorded while simultaneously Calcium Imaging is used to measure odor evoked activity in mushroom body neurons in vivo.

Detection of Phospholipase C Activity in the Brain Homogenate from the Honeybee

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.
