Lamin Antibody Staining

Lamin antibody staining is an immunostaining method used to detect lamins, structural proteins that form the nuclear lamina and help maintain nuclear shape, organization, and mechanical stability. In fixed and permeabilized cells or tissue, primary antibodies bind specific lamin proteins, while labeled secondary antibodies produce a fluorescent or enzymatic signal that reveals their distribution by microscopy. In neuroscience, this approach helps characterize nuclear architecture in neurons and glial cells, track changes during development or differentiation, and investigate how altered lamin expression or organization relates to cellular stress and neurological disease. The resulting spatial information supports studies of nuclear function and brain pathology.

This protocol presents steps taken to dissect ovaries in the freshwater planarians, Schmidtea mediterranea. The dissected ovaries are compatible for antibody immunostaining and ultrastructural analysis with transmission electron microscopy to study the cell biology of the oocytes and somatic cells, providing an imaging depth and quality that were previously inaccessible.

We describe a streamlined protocol for generating "fillet" preparations of Drosophila embryos of specific genotypes. This protocol allows efficient execution of a variety of genetic screens. It also allows excellent visualization of structures in the late embryo.
