Leukocyte Adhesion

Leukocyte adhesion is the multistep process by which white blood cells attach to vascular endothelium and leave the bloodstream to reach sites of infection, injury, or inflammation. Inflammatory signals induce endothelial cells to display adhesion molecules; selectins mediate initial leukocyte rolling, while chemokines activate integrins that bind endothelial ligands and produce firm adhesion before transendothelial migration. In neuroscience, this process helps regulate immune-cell entry into nervous tissue and can influence neuroinflammation, blood-brain barrier function, and responses to injury or disease. Understanding leukocyte adhesion supports research into how immune and vascular mechanisms contribute to neurological disorders and potential therapeutic targets.

Imaging Leukocyte Adhesion to the Vascular Endothelium at High Intraluminal Pressure

This is a method to visualise leukocyte adhesion to the endothelium in harvested pressurised vessels. The technique enables studying vascular adhesion under shear flow with differing intraluminal pressures up to 200 mmHg thus mimic-ing the pathophysiological conditions of high blood pressure.

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.
