Linear Array Recording

Linear array recording is a neuroscience technique that uses multiple electrode contacts arranged along a straight shank to measure electrical activity at different depths within neural tissue. As the array is positioned through a brain region, each contact simultaneously detects extracellular voltage fluctuations, including action potentials and local field potentials, across cortical layers or other anatomical structures. Comparing signals along the array reveals how neural activity varies spatially and propagates through tissue, while depth-resolved recordings can help identify layer-specific responses and circuit interactions. This approach supports studies of sensory processing, network dynamics, brain connectivity, and the physiological effects of stimulation or disease.

In this protocol we demonstrate how to fabricate and condition tetrodes for use with a micro-drive array, which was designed for chronic electrophysiological recordings in rats. In addition, we illustrate the final stages of micro-drive array construction, which includes installing ground wires and a protective cone.

A robust way to study neuronal avalanches, i.e. scale-invariant spatio-temporal activity bursts, indicative of critical state dynamics in cortex. Avalanches emerge spontaneously in developing superficial layers of cultured cortex which allows for long-term measurements of the activity with planar integrated multi-electrode arrays (MEA) under precisely controlled conditions.

This protocol provides the necessary information for setting up, caring for, recording from and electrically stimulating cultures on MEAs. In vitro networks provide a means for asking physiologically relevant questions at the network and cellular levels leading to a better understanding of brain function and dysfunction.
