Mitochondrial Labeling

Mitochondrial labeling is the use of fluorescent, genetic, or chemical markers to identify and track mitochondria in cells, revealing their distribution, movement, and structural changes. Labels work by selectively accumulating within mitochondria through membrane-potential-dependent dyes, binding mitochondrial components, or directing fluorescent proteins to the organelle with targeting sequences; microscopy then records labeled organelles over time. In neuroscience, these approaches help researchers examine mitochondrial transport along axons and dendrites, energy production at synapses, and changes associated with neuronal injury or disease. Quantifying mitochondrial number, morphology, and dynamics can connect organelle behavior with neuronal function and guide studies of neurodegeneration and brain development.

In Situ Labeling of Mitochondrial DNA Replication in Drosophila Adult Ovaries by EdU Staining

Drosophila oogenesis continues to be exceptionally useful in the study of mitochondrial proliferation and inheritance. This manuscript describes a detailed protocol used to label the replicating mitochondrial DNA (mtDNA) in Drosophila adult ovaries with 5-ethynyl-2´-deoxyuridine (EdU), which facilitates uncovering mechanisms associated with mitochondrial inheritance that were previously debatable.

Visualization of Mitochondrial Respiratory Function using Cytochrome C Oxidase / Succinate Dehydrogenase (COX/SDH) Double-labeling Histochemistry

The cytochrome c oxidase/sodium dehydrogenase (COX/SDH) double-labeling method allows for direct visualization of mitochondrial respiratory enzyme deficiencies in fresh-frozen tissue sections. This is a straightforward histochemical technique and is useful in investigating mitochondrial diseases, aging, and aging-related disorders.

This protocol describes a procedure to study the respiration of mitochondria isolated from skeletal muscles. This method was adapted from Scorrano et al. (2007). The mitochondrial isolation procedure requires about 2 hours. The mitochondrial respiration can be completed in about 1 hour.
