Mouse Brain Connectivity

Mouse brain connectivity is the study of how neurons and brain regions are linked through anatomical projections and functional interactions, providing a map of circuits that support behavior and cognition. Researchers characterize these networks using methods such as viral or fluorescent tracer tracing, genetic labeling, electrophysiology, and imaging to follow axons, identify synaptic partners, and measure activity across connected regions. In neuroscience, connectivity maps help relate circuit organization to sensory processing, learning, movement, and disease-related changes. Because the mouse brain can be studied with cell-type-specific tools and controlled experiments, these data support mechanistic models of neural function and comparisons with human brain disorders.

Resting-state functional-connectivity MRI has identified abnormalities in patients with a wide range of neuropsychiatric disorders, including epilepsy due to malformations of cortical development. Transcranial Magnetic Stimulation in combination with EEG can demonstrate that patients with epilepsy have cortical hyperexcitability in regions with abnormal connectivity.

We describe a protocol allowing the purification of the mouse brain's vascular compartment. Isolated brain vessels include endothelial cells linked by tight junctions and surrounded by a continuous basal lamina, pericytes, vascular smooth muscle cells, as well as perivascular astroglial membranes.
