Multi-laboratory Stroke

Stroke is a neurological emergency in which disrupted cerebral blood flow, caused by vessel blockage or bleeding, damages brain tissue and can impair movement, speech, cognition, or sensation. Multi-laboratory stroke research coordinates experiments across independent institutions, using harmonized protocols, shared outcome measures, and comparable models or clinical data to test whether findings are reproducible. This approach strengthens confidence in mechanisms of neuronal injury and recovery while revealing how biological variability, experimental conditions, and treatment timing influence results. In neuroscience, collaborative stroke studies support more reliable biomarker discovery, therapy development, and translation of preclinical findings into clinically relevant interventions.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

Various murine models of middle cerebral artery occlusion (MCAO) are widely used in experimental brain research. Here, we demonstrate the model of transcranial permanent distal MCAO which produces consistent cortical infarction of a size corresponding to damage imposed by the majority of human ischemic strokes.
